Rett syndrome Rett syndrome is a rare genetic disorder that primarily affects girls and women, causing a progressive loss of motor and cognitive skills. Until now, there was no effective treatment for this devastating disease. However, today we have encouraging news: the drug trofinetide has proven to be safe and effective in treating Rett syndrome, according to data from the LAVANDER study, recently published in the prestigious journal Nature Medicine.
This study, based on data from a phase 3 clinical trial, establishes trofinetide as the first approved drug for the treatment of Rett syndrome. Two previous phase 2 trials had already shown the potential efficacy of this synthetic drug, similar to the naturally occurring glycine-proline-glutamate growth factor found in the brain, in reducing symptoms in small groups of patients with Rett syndrome.
The LAVANDER study, coordinated by James Youakim's team at Acadia Pharmaceuticals, randomly assigned 187 female patients with Rett syndrome to receive either trofinetide or a placebo for 12 weeks. To assess treatment efficacy, the Rett Syndrome Behavior Questionnaire (RSBQ) score and the Clinical Global Impression-Improvement (CGI-I) scale, a global assessment performed by physicians to determine patient improvement or worsening, were used.
The results were promising: significant improvements were observed in both RSBQ and CGI-I scores in the trofinetide group, with no reports of serious treatment-related adverse effects. This suggests that trofinetide is relatively well tolerated and shows efficacy in improving key signs and symptoms associated with Rett syndrome.
As a result of these findings, the U.S. Food and Drug Administration (FDA) has approved trofinetide for the treatment of Rett syndrome in adult and pediatric patients. Since late April 2023, the drug, marketed as DAYBUE™, has been available in the United States. However, its submission to European health authorities has not yet been announced, so its availability in other countries is pending.
This breakthrough represents significant hope for people living with Rett syndrome and their families. Trofinetide offers an opportunity to improve the quality of life for these individuals by alleviating symptoms and helping them maintain enhanced functionality. Furthermore, this milestone highlights the importance of research and development of specialized treatments for rare disorders, demonstrating that medical science continues to advance toward innovative and effective solutions.
